DNA Sequencing Team
High-Throughput Sequencing Unit

The primary mission of the High-Throughput Sequencing and Genotyping Unit is to facilitate the unraveling of genetic information across the large and diverse collection of animals, plants and microbes that are the research focus of many of our faculty. The unit prepares libraries and generates DNA templates and sequence data using Sanger, pyrosequencing ( Roche/454 GSFLX Titanium), and Illumina Genome Analyzer IIx sequencing platforms. These resources are also critically important to the operations of our sister units, Bioinformatics and Functional Genomics.

In addition, the facility provides DNA services for investigators requiring core DNA sequencing, fragment analysis, and custom oligonucleotide synthesis. All of the unit’s services include consultation and assistance in all aspects of a project. Researchers are encouraged to discuss their projects with facility personnel who would be glad to provide information and quotes for their projects.

Services:

Roche/454 Genome Sequencer FLX-Titanium:

  • Custom cDNA library construction for transcriptome sequencing.
  • Preparation of single-stranded template DNA library (sstDNA) for 454 sequencing from genomic DNA, fosmids, long PCR products, etc.
  • Titrations of 454-adapted libraries.
  • Sequencing on full plate(s) or smaller regions.

Illumina Genome Analyzer IIx:

  • Construction of libraries from genomic DNA, small RNAs, mRNA, CHIP DNA, etc.
  • Sequencing on the GAIIx:
    • full flowcell (7 lanes) or any number of lanes
    • single read
    • paired-ends and mate-pair libraries
    • 36bp, 50bp or 76bp

    Sanger sequencing:

    • Custom cDNA and genomic library construction.
    • Colony picking (QPixII).
    • Plasmid preps (heat lysis or Qiagen 9600 BioRobot).
    • Sequencing on ABI 3730xl capillary systems.
