Question: What are antigen requirements?
Answers:
- Quantity : 1.5 - 2 mg for a new antibody generation with 1 rabbit, 1 chicken, 5 mice or 3 rats.
- Purity : 90% or better. Contact us if an antigen is less than 90% pure. We might help you with further purification or accept an antigen as it is.
- Concentration : minimum 1mg/ml. Accurate determination of antigen concentration is critical for a project success.
- Buffer : Tris or phosphate buffer with a neutral pH (7-8) is acceptable. Detergents, reducing agents and imidazole that are often associated with protein purification should be removed by dialysis or buffer exchange. Most efficient dialysis method for our purpose is to use dialysis tubing with 1-2ml samples in 1L buffer stirring overnight at 4C. If an antigen does not mixed well with adjuvant due to insufficient dialysis (this happens very often!), $25 fee will be charged for further dialysis by us.
- Sterilization : passing through 0.22um filter unit before concentration is determined.
- Shipping: Dry ice or ice pack with overnight service.
- Insoluble antigen : Insoluble antigen is acceptable if concentration is well measured. Contact us if you have problem determining concentration with an insoluble antigen.

Question: What dilution do I use to test an antibody sample? Answers :
Hybridoma medium can be tested without any dilution or dilute as much as 10 x for high affinity antibody.
A serum sample can be diluted 1:200 to 1:10,000. For rabbit serum, first bleed can be tested at 1:500 to 1:5000 dilutions.
Final bleeds to be diluted to 1: 20,000. Mouse serum first bleeds can be tested at 1:200 to 1:1000. Later bleeds can be tested up to 1:5000.
A chicken serum can be tested as a rabbit serum. Chicken egg yolk and chicken serum contain the similar concentration of antibody IgY.
