After mouse ES cell lines having undergone homologous recombination with the targeting vector are identified, chimeric mice can be generated by microinjection of those ES cells into 3.5 day mouse embryos. The facility will perform the microinjection and transfer injected blastocysts into the uteri of foster females. About 3 weeks after pups are born, the facility will transfer all the chimeric mice to investigator’s protocol.

To request service from Transgenic Mouse Facility:
- Provide the facility with the knock out construct approval date from Institutional Biosafety Committee (IBC) and an approved protocol for animal research from IACUC. The facility will not initiate any experiment without the approvals from IBC and IACUC.
- Arrange an appointment with the facility Director, Fuming Pan (244-0649, fpan@illinois.edu) to initiate ES cell microinjection.
- Complete and submit the Service Request Form for knockout mouse production.
- If the investigator already has ES cell clones, please provide the facility with ES cells prepared according to the specified protocol on the day of injection.
- TMF will provide at least 2 chimeric mice per knock out construct.
- All the offspring in the Transgenic Mouse Facility and the cage cost will be transferred to the investigator at the weaning day.
